The standard blues scale is I - b3 - 4 - #4 - 5 - b7 - 8 of a major scale. A C major blues scale would be the notes C - Eb - F - F# - G - Bb - C. This is the most common blues scale and almost always what people mean when they say the "blues scale". It's the combination of notes, the bends, style and inflection that breathes life into blues music!

A "country" version of the blues scale adds the major 6th to the scale. A C major blues scale would then add the "A" for C - Eb - F - F# - G - Bb - C.

Another common addition is the major second or the "D" in the key of C for C - D - Eb - F - F# - G - Bb - C.

The third is part of the "color" of the blues scale, the part that makes it "blue". The MAJOR third can be used on the 1 chord. In other words, if you are in the key of C you can use the note E (not Eb) on the C (1) chord. On the F (4) chord you would switch to the flatted third or Eb.

A standard twelve bar blues progression is:
1 / 1 / 1 / 1
4 / 4 / 1 / 1
5 / 4 / 1 / 1

Now add a "4" chord on the second measure:
1 / 4 / 1 / 1
4 / 4 / 1 / 1
5 / 4 / 1 / 1

Now add a "5" chord on the turnaround:
1 / 4 / 1 / 1
4 / 4 / 1 / 1
5 / 4 / 1 / 5

This last chord progression is the most common blues progression and generally what people mean when they refer to the "12 bar blues" progression.
